Title: Production of anti luteinizing hormone releasing factor (LH-RF) serum: immunological application and development of radioimmunoassay. Author: Shiina M. Abstract: The successful production of an antiserum to LH-RF, development of a radioimmunoassay, and biological experiments with this antiserum are described. This specific antiserum was produced in female rabbits by repeated intradermal injections of LH-RF-bovine serum albumin (BSA) conjugate emulsified in Freund's complete adjuvant. High specificity of the antiserum to LH-RF was shown. By a modified chloramine T method, with shortening of the time of action, radioiodination of LH-RF was achieved. The degradation of exogenous LH-RF in man was shown to be very rapid. The half-life was 3.9 minutes within the first 15 minutes. Serum levels of LH were similar after iv and im injections of LH-RF. Iv injections of anti-LH-RF serum to 4-day cycling female rats on the proestrous day blocked ovulation on the following day. Presumably, endogenous LH-RF was neutralized before it reached the anterior pituitary gland. However, serum LH was maintained at basal levels. Apparently the anterior pituitary can secrete the basal levels of LH without any stimulation by the hypothalamic LH-RF. After 1 year, the 2 female rabbits which had produced the anti-LH-RF sera showed marked atrophy of ovaries and uteri histologically. Their pituitaries showed no marked change. Serum LH and 20alpha-hydroxypregn-4-en-3-one concentrations in these rabbits were low but detectable by radioimmunoassays. Data show that the specific antiserum to LH-RF is useful for biological studies as well as for the development of a radioimmunoassay ofLH-RF.[Abstract] [Full Text] [Related] [New Search]
