    Abstract:
    OBJECTIVE: To establish an analytical method for the fingerprint of triterpenoid constituents of Poria cocos by UPLC-UV-MS and compare the fingerprints of different medicinal parts of Poria cocos. METHODS: With dehydropachymic acid as reference substance, the separation was performed on Shim-pack XR-ODS II (75 mm x 2.0 mm, 2.2 microm) analytical column. The mobile phase was consisted of acetonitrile and 0.1% formic acid as gradient eluent. The UV detection wavelength was 242 nm. The flow rate was 0.5 mL/min. The column temperature was 30 degrees C. The cluster analysis was carried on by SPSS 16. 0. RESULTS: The UPLC-UV-MS fingerprints of triterpenoid constituents of Poria cocos were set up. The result showed that 22 peaks were found in different medicinal parts and 12 peaks of them were common. The results of method validation met technical requirement of fingerprints; Triterpenoid constituents in pared skin of Poria and peeled and sliced Poria were different, and the effect of habitat on the quality of peeled and sliced Poria was more obvious than that of pared skin of Poria. CONCLUSION: The method is stable and reliable with a good reproducibility and provides a reference standard for the quality control of Poria cocos.
