Title: [Moxibustion improves learning-memory ability by regulating apoptosis related proteins of hippocampus in diabetic rats with cognitive impairment]. Abstract: OBJECTIVE: To investigate the effect of moxibustion on the proteins related with apoptosis and nuclear transcription factor kappa B (NF-κB) in hippocampus of diabetic rats with cognitive impairment (CI), so as to explore its mechanism underlying improvement of learning-memory ability. METHODS: Thirty SD rats were randomly divided into normal, model and moxibustion groups (n=10 rats/group). The diabetic model was established by i.p. injection of streptozotocin solution (25 mg·kg-1·d-1), followed by high-fat diet raising for 4 weeks, and the CI model was confirmed by Morris water maze test. The rats in the moxibustion group were given moxibustion at "Shenting" (GV24), "Baihui" (GV20) and "Dazhui" (GV14) for 20 min each time, the treatment was conducted 6 times a week for 4 weeks. The learning-memory ability was detected by Morris water maze test, the random blood glucose of rats was measured by glucometer and test strips. The protein and mRNA expression levels of Bcl-2, Bax, Caspase-3 and NF-κB p65 in hippocampus were detected by Western blot and quantitative real-time PCR, separately. RESULTS: After modeling, the random blood glucose, escape latency, and the expression levels of Bax, Caspase-3 and NF-κB p65 proteins and mRNAs in the model group were significantly increased, while the expression levels of Bcl-2 protein and mRNA were decreased (P<0.001,P<0.01, P<0.05) in comparison with the normal group. Following the treatment, the modeling induced increase of blood glucose, escape latency, and the expression levels of Bax, Caspase-3 and NF-κB p65 proteins and mRNAs, as well as decrease of Bcl-2 protein and mRNA expression levels were reversed (P<0.05, P<0.01, P<0.001). CONCLUSION: Moxibustion can improve learning-memory ability in diabetic rats with cognitive impairment, which may be related to its function in regulating the expression levels of hippocampal Bcl-2, Bax, Caspase-3 and NF-κB. 目的:观察艾灸对糖尿病认知减退大鼠海马凋亡相关蛋白、核转录因子κB(NF-κB)的影响,探讨艾灸改善学习记忆的作用机制。方法:SD大鼠随机分为正常组、模型组和艾灸组,每组10只。以高脂高糖饲料联合小剂量链脲佐菌素腹腔注射诱导糖尿病认知功能减退大鼠模型。艾灸组艾灸“神庭”“百会”及“大椎”,每次20 min,每周6次,治疗4周。检测各组大鼠随机血糖,水迷宫实验测定各组大鼠学习记忆能力,Western blot及实时荧光定量PCR法检测各组大鼠海马B淋巴细胞瘤2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)和NF-κB p65蛋白及mRNA相对表达量。结果:治疗后,模型组大鼠随机血糖值较正常组明显升高(P<0.001),艾灸组大鼠随机血糖值较模型组明显降低(P<0.05)。模型组大鼠逃避潜伏期较正常组明显延长(P<0.01),艾灸组大鼠逃避潜伏期较模型组明显缩短(P<0.01)。模型组大鼠海马Bax、Caspase-3、NF-κB p65蛋白及mRNA表达较正常组明显升高(P<0.001),Bcl-2蛋白及mRNA表达较正常组明显降低(P<0.001,P<0.05),艾灸组大鼠海马Bax、Caspase-3和NF-κB p65蛋白及mRNA表达较模型组明显降低(P<0.001),Bcl-2蛋白及mRNA表达较模型组明显升高(P<0.001,P<0.05)。结论:艾灸可以调控糖尿病认知减退大鼠海马凋亡相关蛋白表达,改善认知功能,NF-κB信号通路可能参与其中。.[Abstract] [Full Text] [Related] [New Search]
