    Abstract:
    The effect of estradiol infusion on uterine activity and peripheral levels of prostaglandin F and progesterone in pregnant women at term were investigated. 5 normal women at term and 4 women with pre-eclampsia at term were given estradiol-17 beta by infusion over 2-4 hours. Radioimmunoassay was used to measure peripheral plasma levels of estradiol, prostaglandin F, and progesterone. During the estradiol infusion, 8 of the 9 patients developed mildly painful uterine contractions which lasted 30-40 seconds and occurred at 3-7 minute intervals. Estradiol levels increased appropriately beyond base line. There was no significant change in progesterone or prostaglan F levels in any patient, but all patients except 1 developed an increase in uterine activity. Results of this experiment as well as the bulk of available evidence suggest that peripheral levels of progesterone do not decrease unless there has been a significant increase in prostaglandin F2alpha, significant placental disruption, or uterine contractions sufficient to constitute active labor.
