  • Title: [Recombination in the plasmid transformation of Bacillus subtilis: the role of superhelical DNA].
    Author: Bashkirov VI, Prozorov AA.
    Journal: Genetika; 1983; 19(2):204-10. PubMed ID: 6403406.
    Abstract:
The inhibitor of DNA gyrase, novobiocine affects recombination between homologous plasmids in the course of plasmid transformation. Novobiocine decreased significantly the yield of recombinants when added to competent cells during the first 10 minutes. Later, after 30 to 40 min, addition of the antibiotic had no effect. Probably, recombination is completed, in general, in 10 to 20 min after DNA uptake. Plasmid recombination was depressed totally after preincubation of competent cells with the drug. Evidently, inhibition of recombination was due to the loss of superhelicity in resident plasmid DNA.
