Title: Inhibition of Tamoxifen of the stimulatory action of FSH on oestradiol-17beta synthesis by rat ovaries in vitro. Author: Watson J, Howson JW. Journal: J Reprod Fertil; 1977 Mar; 49(2):375-6. PubMed ID: 850224. Abstract: Inhibition by tamoxifen of the stimulatory action of follicle stimulating hormone (FSH) on estradiol-17-beta synthesis by rat ovaries in vitro was investigated. Ovaries were removed at Day 4 of pregnancy or at various stages of the estrous cycle. Tissue incubations were carried out for 3 hours at 37 degrees C in 3 ml of Medium 199 containing radioactive androstenedione or testosterone. 15 mcg tamoxifen and 10 mcg FSH were tested together or separately. With ovarian tissue from pregnant rats FSH caused a significant increase (p less than .01) in the incorporation of androstenedione or testosterone into estradiol relative to control incubations while tamoxifen caused significant (p less than .01) decreases. When FSH and tamoxifen were present the stimulatory action of FSH was completely abolished and the incorporation of both precursors alone appeared to be slightly less than that seen with tamoxifen alone.
